A mutation in the general membrane trafficking machinery and hydrocephaly.

چکیده
M embrane fusion is required to deliver specific cargo molecules to distinct intracellular compartments, thereby maintaining the functional and structural organization of eukaryotic cells, as well as intercellular communication, which occurs in precisely timed spatial patterns at the plasma membrane. During embryogenesis, exocytosis is harnessed to induce differentiation and morphogenesis, as demonstrated by the work of Hong et al. in this issue of PNAS (1). Later and throughout life, the temporal control of site-specific fusion coordinates the release and processing of short and long-range messages within an organism.

The gene for soluble N-ethylmaleimide sensitive factor attachment protein alpha is mutated in hydrocephaly with hop gait (hyh) mice.
The spontaneous autosomal recessive mouse mutant for hydrocephaly with hop gait (hyh) exhibits dramatic cystic dilation of the ventricles at birth and invariably develops hopping gait. We show that the gene for soluble N-ethylmaleimide sensitive factor attachment protein alpha, also known as alpha-SNAP, is mutated in hyh mice. alpha-SNAP plays a key role in a wide variety of membrane fusion eve...

Abstract The S362A mutation block ROMK2 (Kir1.1b) endocytosis in Xenopus laevis oocyte membrane . Saeed Hajihashemi1 , 1-Assistant professor, PhD in Physiology, Department of Physiology, School of Medical science, Arak University of Medical Sciences. Introduction: ROMK channel is localized on the apical membrane of the nephron. Recent studies suggest that endocytosis of ROMK chan...
متن کاملRab11 in Disease Progression
Membrane/ protein trafficking in the secretory/ biosynthetic and endocytic pathways is mediated by vesicles. Vesicle trafficking in eukaryotes is regulated by a class of small monomeric GTPases the Rab protein family. Rab proteins represent the largest branch of the Ras superfamily GTPases, and have been concerned in a variety of intracellular vesicle trafficking and different intracellular sig...
متن کاملCharcot-Marie-Tooth disease-linked protein SIMPLE functions with the ESCRT machinery in endosomal trafficking
Mutations in small integral membrane protein of lysosome/late endosome (SIMPLE) cause autosomal dominant, Charcot-Marie-Tooth disease (CMT) type 1C. The cellular function of SIMPLE is unknown and the pathogenic mechanism of SIMPLE mutations remains elusive. Here, we report that SIMPLE interacted and colocalized with endosomal sorting complex required for transport (ESCRT) components STAM1, Hrs,...
متن کاملWhen intracellular logistics fails--genetic defects in membrane trafficking.
The number of human genetic disorders shown to be due to defects in membrane trafficking has greatly increased during the past five years. Defects have been identified in components involved in sorting of cargo into transport carriers, vesicle budding and scission, movement of vesicles along cytoskeletal tracks, as well as in vesicle tethering, docking and fusion at the target membrane. The ner...
